Lion’s Mane, erinacines, NGF and cognition: from mycelium to a small trial
Trace Lion’s Mane research from erinacine chemistry and nerve-growth-factor experiments to the 30-person mild-cognitive-impairment trial.
Erinacines were isolated from mycelia in laboratory work. A small 30-person trial used dried mushroom tablets, not an Enki tincture, and scores declined after stopping.
Compounds & molecular formulae
Erinacine A
C₂₅H₃₆O₆
A cyathane diterpenoid isolated from Hericium mycelia and studied for NGF-related activity.
PubChem compound record ↗β-Glucan repeat
(C₆H₁₀O₅)ₙ
A shorthand for polysaccharide repeat units; it does not identify branching, molecular weight or extract dose.
PubChem compound record ↗The proposed NGF route
- Mycelial or fruiting-body compounds are extracted
- An experimental model changes NGF expression
- NGF engages TrkA-related signalling
- Human cognition must be tested with validated scales
NGF pathway diagrams are preclinical evidence until a comparable human product and endpoint are studied [1][2].
Mycelium and fruiting body are not interchangeable
Hericium erinaceus contains different compound profiles in fruiting bodies, cultured mycelia and extraction fractions. Kawagishi and colleagues reported erinacines A, B and C as strong stimulators of NGF synthesis from cultured mycelia [1]. That is an important discovery, but an erinacine result cannot be assumed for a fruiting-body powder or a tincture unless the material and assay are documented. “Lion’s Mane” is a species name, not a certificate of erinacine content.
The small human MCI trial
Mori and colleagues randomised 30 adults aged 50–80 with mild cognitive impairment to four 250 mg tablets containing 96% dry Yamabushitake powder three times daily or placebo for 16 weeks [2]. Scores on a cognitive scale were higher in the mushroom group at weeks 8, 12 and 16, then fell significantly four weeks after intake stopped. The study was small and short; it used a defined dry powder and did not establish prevention of dementia, nerve regeneration or a tincture equivalence.
How NGF and TrkA fit together
NGF binds TrkA and p75 neurotrophin receptors, activating pathways such as PI3K–AKT, RAS–RAF–MEK–ERK and PLC-γ that influence neuronal survival, differentiation and plasticity. In a dish, increased NGF or neurite growth can be measured. In a person, cognition depends on sleep, vascular health, medications, mood, hearing, education and many other factors. A mechanistic pathway is a reason to study a compound—not a reason to promise memory restoration.
Questions for a tincture buyer
Ask whether the product uses fruiting body, mycelium or both; the extraction solvent; extract ratio; alcohol content; beta-glucan or erinacine testing; serving information; and allergen controls. If you have a mushroom allergy, are pregnant or take medicines, ask a clinician. Enki’s product page can provide current label information, but the research above should not be read as a clinical prescription.
